Viruses are small infectious agents that can replicate only inside the living cells of an organism. Viruses infect all forms of life, including animals, plants, fungi, bacteria and archaea. They are found in almost every ecosystem on Earth and are the most abundant type of biological entity, with millions of different types, although only about 6,000 viruses have been described in detail. Some viruses cause disease in humans, and others are responsible for economically important diseases of livestock and crops.
Virus particles (known as virions) consist of genetic material, which can be either DNA or RNA, wrapped in a protein coat called the capsid; some viruses also have an outer lipid envelope. The capsid can take simple helical or icosahedral forms, or more complex structures. The average virus is about 1/100 the size of the average bacterium, and most are too small to be seen directly with an optical microscope.
The origins of viruses are unclear: some may have evolved from plasmids, others from bacteria. Viruses are sometimes considered to be a life form, because they carry genetic material, reproduce and evolve through natural selection. However they lack key characteristics (such as cell structure) that are generally considered necessary to count as life. Because they possess some but not all such qualities, viruses have been described as "organisms at the edge of life".
Rabies is a disease of humans and some other mammals, generally caused by the rabies virus, an RNA virus in the Rhabdoviridae family. A few cases have involved the closely related Australian bat lyssavirus. Rabies virus has a wide host range. Globally, dogs are the main source of human infections, with bats being important in the Americas; other naturally infected animals include monkeys, raccoons, foxes, skunks, cattle, horses, wolves, coyotes, cats, mongooses, bears, groundhogs, weasels and other carnivores. Transmission is commonly via saliva, usually but not always from bites; it can potentially occur via aerosols contacting mucous membranes. The typical human incubation period is 1–3 months. The neurotropic virus travels along neural pathways into the CNS and brain, where it causes meningoencephalitis. Nonspecific symptoms such as fever and headache are followed by neurological symptoms, including partial paralysis, confusion, agitation, paranoia, hallucinations and sometimes hydrophobia, which progress to delirium, coma and death. Around 17,400 people died from rabies in 2015, mainly in Asia and Africa.
Rabies is mentioned in the Codex of Eshnunna of around 1930 BC. The first vaccine was developed in 1885 by Louis Pasteur and Émile Roux. Prophylactic vaccination is used in people at high risk, pets and wild animals. Post-exposure prophylaxis, including vaccine and immunoglobulin, is completely effective if begun immediately after exposure, but survival is extremely rare once symptoms have begun.
The castor bean tick, Ixodes ricinus, can transmit the tick-borne encephalitis virus. Ticks are common vectors for viruses, and other tick-borne diseases include Colorado tick fever and Crimean–Congo haemorrhagic fever.

A prion is an infectious agent hypothesised to consist of protein. This is in contrast to viruses and other known infectious agents, which all contain one or both of the nucleic acids, DNA and RNA. Prions propagate by transmitting a misfolded protein state. The prion induces existing, properly folded proteins in the host to convert into the misfolded prion form. Abnormal protein aggregates called amyloids accumulate in infected tissue and are associated with tissue damage and cell death.
Prion variants of PrP are associated with the transmissible spongiform encephalopathies in mammals. Human prion diseases include Creutzfeldt–Jakob disease, Gerstmann–Sträussler–Scheinker syndrome, fatal familial insomnia, kuru and variably protease-sensitive prionopathy. Prion diseases of other mammals include bovine spongiform encephalopathy ("mad cow disease") in cattle, scrapie in sheep, and chronic wasting disease in deer. All known mammalian prion diseases affect the structure of the brain or other neural tissue. All are progressive, lack an effective treatment and are inevitably fatal. Proteins showing prion-type behaviour are also found in some fungi. Fungal prions do not appear to cause disease in their hosts.
The last recorded smallpox death occurred during the 1978 smallpox outbreak in Birmingham, UK. The outbreak resulted from accidental exposure to the Abid strain of Variola major, from a laboratory, headed by Henry Bedson, at the University of Birmingham Medical School – also associated with an outbreak in 1966. Bedson was investigating strains of smallpox known as whitepox, considered a potential threat to the smallpox eradication campaign, then in its final stages.
A medical photographer who worked on the floor above the laboratory showed smallpox symptoms in August and died the following month; one of her contacts was also infected but survived. The government inquiry into the outbreak concluded that she had been infected in late July, possibly via ducting, although the precise route of transmission was subsequently challenged. The inquiry criticised the university's safety procedures. Bedson committed suicide while under quarantine. Radical changes in UK research practices for handling dangerous pathogens followed, and all known stocks of smallpox virus were concentrated in two laboratories.

Tobacco mosaic virus (TMV) is an RNA virus in the Virgaviridae family that infects a wide range of plants, including tobacco, tomato, pepper, other members of the Solanaceae family, and cucumber. The rod-shaped virus particle is around 300 nm long and 18 nm in diameter, and consists of a helical capsid made from 2130 copies of a single coat protein, which is wrapped around a positive-sense single-stranded RNA genome of around 6400 bases. The coat protein and RNA can self-assemble to produce infectious virus.
Infection often causes characteristic patterns, such as "mosaic"-like mottling and discoloration on the leaves, but is almost symptomless in some host species. TMV causes an economically important disease in tobacco plants. Transmission is frequently by human handling, and prevention of infection involves destroying infected plants, hand washing and crop rotation to avoid contaminated soil. TMV is one of the most stable viruses known. The fact that it does not infect animals and can readily be produced in gramme amounts has led to its use in numerous pioneering studies in virology and structural biology. TMV was the first virus to be discovered and the first to be crystallised.
Sir Frank Macfarlane Burnet (3 September 1899 – 31 August 1985) was an Australian virologist, microbiologist and immunologist. His early virological studies were on bacteriophages, including the pioneering observation that bacteriophages could exist as a stable non-infectious form that multiplies with the bacterial host, later termed the lysogenic cycle.
With the outbreak of World War II, Burnet's focus moved to influenza. Although his efforts to develop a live vaccine proved unsuccessful, he developed assays for the isolation, culture and detection of influenza virus, including haemagglutination assays. Modern methods for producing influenza vaccines are still based on his work improving virus-growing processes in hen's eggs. He also researched influenza virus genetics, examining the genetic control of virulence and demonstrating, several years before influenza virus was shown to have a segmented genome, that the virus recombined at high frequency.

Ribavirin is a nucleoside analogue that mimics the nucleoside guanosine. It shows some activity against a broad range of DNA and RNA viruses, but is less effective against dengue fever, yellow fever and other flaviviruses. The drug was first synthesised in the early 1970s by Joseph T. Witkowski and Roland K. Robins. Ribavirin's main current use is against hepatitis C, in combination with pegylated interferon, nucleotide analogues and protease inhibitors. It has been used in the past in an aerosol formulation against respiratory syncytial virus-related diseases in children. Ribavirin has been used in combination as part of an experimental treatment for rabies. It is also the only available treatment for the viruses causing some viral haemorrhagic fevers, including Lassa fever, Crimean–Congo haemorrhagic fever and hantavirus disease, but is ineffective against the filovirus diseases, Ebola and Marburg. Clinical use is limited by the drug building up in red blood cells to cause haemolytic anaemia.
